To better understand the functional mechanism of four types of tea (green tea, black tea, jasmine tea, and dark tea) on the quality of stewed beef, changes in quality characteristics, proteomics, and metabolomics were investigated. Adding these four tea types decreased the pH value, L* value, shear force, and hardness of the stewed beef. Among these groups, black tea (BT) significantly improved the tenderness of the stewed beef. They have substantially impacted pathways related to protein oxidative phosphorylation, fatty acid degradation, amino acid degradation, and peroxisomes in stewed beef. The study identified that Myosin-2, Starch binding domain 1, Heat shock protein beta-6, and Myosin heavy chain four are significantly correlated with the quality characteristics of tea-treated stewed beef, making them potential biomarkers. Green tea (GT), black tea (BT), jasmine tea (JT), and dark tea (DT) led to the downregulation of 20, 36, 38, and 31 metabolites, respectively, which are lipids and lipid-like molecules in the stewed beef. The co-analysis of proteomics and metabolomics revealed that differential proteins significantly impacted metabolites associated with carbohydrates, amino acids, lipids, and other nutrients. This study determined the effects of four types of tea on the quality of stewed beef and their underlying mechanisms, providing valuable insights for applying of tea in meat products. At the same time, it can offer new ideas for developing fresh meat products.

Introduction: Limb-salvage surgery has become the mainstream approaches for the treatment of sarcoma in the lower extremity. In cases where the sarcoma infiltrates the primary vessel, concurrent resection of the vessels and vascular reconstruction are required to ensure sufficient resection and preservation of limb function. The objective of this study is to assess the clinical outcomes of patients who underwent vascular reconstruction utilizing synthetic grafts for limb salvage, specifically in terms of postoperative complications and limb functional status. Methods: Between September 2016 and October 2021, 15 consecutive patients who underwent 15 arterial and 3 venous reconstruction procedures were included in this retrospective study. Incidence of postoperative morbidity, graft patency, rate of limb salvage, and overall survival of patients were analyzed. Results: The median follow-up was 12.5 months (range, 4.5-72.0). Graft thrombosis occurred in 5 patients (33.3%) and graft occlusion occurred in 3 patients (20.0%). The median overall survival was 28.0 months with the estimated 2-year and 5-year overall survival of 57.8% and 43.4% respectively. The 1-year and 2-year estimated patency rates of arterial reconstructions were 82.3% and 62.1%, respectively. None of the included patients with limb amputation were observed as a consequence of severe vascular complications, while two patients underwent amputation due to the repeat recurrence, resulting in a limb salvage rate of 86.7%. Conclusion: Our results show that the combination of vascular reconstruction and oncologic resection is a feasible option for preserving limbs in cases of musculoskeletal sarcoma with vessel involvement in the lower extremity. When vascular reconstruction surgery is performed, synthetic substitutes can be effectively used with low perioperative morbidity and an acceptable rate of limb salvage.

Door-to-balloon (DTB) time significantly affects the prognosis of patients with ST-segment elevation myocardial infarction (STEMI). The effects of temporal differences in emergency department (ED) arrival time on DTB time and on different segments of DTB time remain inconclusive. Therefore, we performed a retrospective study in a tertiary hospital between January 2013 and December 2021 and investigated the relationship between a patient's arrival time and both their DTB time and different segments of their DTB time. Of 732 STEMI patients, 327 arrived during the daytime (08:01-16:00), 268 during the evening (16:01-24:00), and 137 at night (00:01-08:00). Significantly higher odds of delay in DTB time were observed during the nighttime (adjusted odds ratio (aOR): 2.87; 95% confidence interval (CI): 1.50-5.51, p = 0.002) than during the daytime. This delay was mainly attributed to a delay in cardiac catheterization laboratory (cath lab) activation-to-arrival time (aOR: 6.25; 95% CI: 3.75-10.40, p < 0.001), particularly during the 00:00-04:00 time range. Age, sex, triage level, and whether patients arrived during the COVID-19 pandemic also had independent effects on different segments of DTB time. Further studies are required to investigate the root causes of delay in DTB time and to develop specific strategies for improvement.

Nuclear targeted delivery has great potential in improving the efficiency of non-viral carrier mediated genome editing. However, direct and efficient delivery of CRISPR/Cas9 plasmid into the nucleus remains a challenge. In this study, a nuclear targeted gene delivery platform based on fluorescent carbon quantum dots (CQDs) was developed. Polyethylenimine (PEI) and polyethylene glycol (PEG) synergistically passivated the surface of CQDs, providing an excitation-independent green-emitting fluorescent CQDs-PEI-PEG conjugate (CQDs-PP) with an ultra-small size and positive surface charge. Here we show that CQDs-PP could bind CRISPR/Cas9 plasmid to form a nano-complex by electrostatic attraction, which can bypass lysosomes and enter the nucleus by passive diffusion, and thereby improve the transfection efficiency. Also, CQDs-PP could deliver CRISPR/Cas9 plasmid into HeLa cells, resulting in the insertion/deletion mutation of the target EFHD1 gene. More importantly, CQDs-PP exhibited a considerably higher gene editing efficiency as well as comparable or lower cytotoxicity relative to Lipo2000 and PEI-passivated CQDs-PEI (CQDs-P). Thus, the nuclear-targeted CQDs-PP is expected to constitute an efficient CRISPR/Cas9 delivery carrier in vitro with imaging-trackable ability.

Nowadays, Transit-Oriented Development (TOD) plays a vital role for public transport planners in developing potential city facilities. Knowing the necessity of this concept indicates that TOD effective parameters such as network accessibility (node value) and station-area land use (place value) should be considered in city development projects. To manage the coordination between these two factors, we need to consider ridership and peak and off-peak hours as essential enablers in our investigations. To aim this, we conducted our research on Chengdu rail-transit stations as a case study to propose our Node-Place-Ridership-Time (NPRT) model. We applied the Multiple Linear Regression (MLR) to examine the impacts of node value and place value on ridership. Finally, K-Means and Cube Methods were used to classify the stations based on the NPRT model results. This research indicates that our NPRT model could provide accurate results compared with the previous models to evaluate rail-transit stations.

The reliable mechanical properties of ceramizable silicone rubber composites during pyrolysis are necessary for their application in the fire-resistant fields. The effects of liquid-phase amount on the mechanical properties of silicone rubber composites are investigated. The results show a positive correlation between the liquid-phase amount and the flexural strength of the residual products pyrolysis below 800 °C. The nano-γ-Al2O3 in the fillers reacts with liquid B2O3 to form aluminum borate above 800 °C, which consumes the liquid phase and strengthens the residual products to a certain extent. Increasing the B2O3 addition and introducing nano-γ-Al2O3 can control the liquid-phase amount in the range of 15% to 30%, which makes the composites have better residual strength and support performance. The residual strength of composites pyrolysis at 500 °C to 1000 °C is higher than 2.50 MPa, and the maximum is up to 18.7 MPa at 1000 °C.

Fiber-reinforced polymers (FRPs) retrofit reinforced concrete (RC) structures. ABAQUS finite element software was used to perform numerical parametric analysis on a group of RC beams in this research. All specimens were retrofitted by FRP strips as an external retrofitting and experimentally tested up to previous researchers' failure points. The range of subjects examined in these RC beams included cracking torque, ultimate torque, angle of twist, and the effect of using FRP on these subjects. We applied artificial neural networks (ANNs) to predict the structural behavior of RC beams under combined torsion and bending to develop the research accuracy. After testing, the ANN results were compared with the ABAQUS results. Consequently, a reasonable examination of the determined mathematical and trial results confirmed this study's logical accuracy in predicting retrofitted RC beams' structural behavior under combined loading.

BACKGROUND: This study explored the feasibility of radiofrequency (RF)-based radiomics analysis techniques for the preoperative prediction of programmed cell death protein 1 (PD-1) in patients with hepatocellular carcinoma (HCC). METHODS: The RF-based radiomics analysis method used ultrasound multifeature maps calculated from the RF signals of HCC patients, including direct energy attenuation (DEA) feature map, skewness of spectrum difference (SSD) feature map, and noncentrality parameter S of the Rician distribution (NRD) feature map. From each of the above ultrasound maps, 345 high-throughput radiomics features were extracted. Then, the useful radiomics features were selected by the sparse representation method and input into support vector machine (SVM) classifier for PD-1 prediction. RESULTS AND CONCLUSION: Among all the RF-based prediction models and the ultrasound grayscale comparative model, the RF-based model using all of the three ultrasound feature maps had the highest prediction accuracy (ACC) and area under the curve (AUC), which were 92.5% and 94.23%, respectively. The method proposed in this paper is effective for the meaningful feature extraction of RF signals and can effectively predict PD-1 in patients with HCC.

OBJECTIVE: To prospectively investigate the feasibility of shear wave elastography (SWE) as a new quantitative and objective method for evaluating the stiffness of the gastrocnemius medialis (GM) muscle during passive stretching in patients with Parkinson's disease (PD). MATERIALS AND METHODS: SWE of the GM muscle was performed in 28 patients with PD [13 female and 15 male; mean age ± standard deviation (SD): 63.0 ± 8.5 years] and 12 healthy controls (5 female and 7 male; mean age ± SD: 59.3 ± 6.4 years) during passive ankle rotation. A Young's modulus-ankle angle curve was constructed. The GM slack angle and baseline Young's modulus (E0) were compared between the markedly symptomatic and mildly symptomatic sides of patients with PD, and healthy controls. Additionally, the correlation between the GM slack angle and the severity of rigidity, and the observer reproducibility of SWE in determining the GM slack angle were evaluated. RESULTS: The GM slack angle was smaller on both the markedly and mildly symptomatic sides in patients with PD than in healthy controls (mean ± SD of -29.13° ± 3.79° and -25.65° ± 3.39°, respectively, vs. -21.22° ± 3.52°; p < 0.001 and p = 0.006, respectively). Additionally, in patients with PD, the GM slack angle on the markedly symptomatic side was smaller than that on the mildly symptomatic side (p = 0.003). The E0 value was lower on both the markedly and mildly symptomatic sides in patients with PD than in healthy controls (mean ± SD of 10.11 ± 2.85 kPa and 10.08 ± 1.88 kPa, respectively, vs. 12.23 ± 1.02 kPa; p = 0.012 and p < 0.001, respectively). However, no significant difference was found between the markedly and mildly symptomatic sides in patients with PD (p = 0.634). A negative linear relationship was observed between the GM slack angle and lower limb rigidity score on the markedly symptomatic side in patients with PD (r = -0.719; p < 0.001). The intraclass correlation coefficients for observer reproducibility of SWE ranged from 0.880 to 0.951. CONCLUSION: The slack angle determined by SWE may be a useful quantitative and reproducible method for evaluating muscle stiffness in patients with PD.

We investigated the use of an Artificial Neural Network (ANN) to predict the Local Bond Stress (LBS) between Ultra-High-Performance Concrete (UHPC) and steel bars, in order to evaluate the accuracy of our LBS equation, proposed by Multiple Linear Regression (MLR). The experimental and numerical LBS results of specimens, based on RILEM standards and using pullout tests, were assessed by the ANN algorithm using the TensorFlow platform. For each specimen, steel bar diameters ([Formula: see text] of 12, 14, 16, 18, and 20, concrete compressive strength ([Formula: see text]), bond lengths ([Formula: see text]), and concrete covers ([Formula: see text]) of [Formula: see text], [Formula: see text], [Formula: see text] and [Formula: see text] were used as input parameters for our ANN. To obtain an accurate LBS equation, we first modified the existing formula, then used MLR to establish a new LBS equation. Finally, we applied ANN to verify our new proposed equation. The numerical pullout test values from ABAQUS and experimental results from our laboratory were compared with the proposed LBS equation and ANN algorithm results. The results confirmed that our LBS equation is logically accurate and that there is a strong agreement between the experimental, numerical, theoretical, and the predicted LBS values. Moreover, the ANN algorithm proved the precision of our proposed LBS equation.

Background: Emerging evidence suggests that gut microbiota plays a vital role in the occurrence of multiple endocrine disorders including polycystic ovary syndrome (PCOS). Shaoyao-Gancao Decoction (SGD), a classical Chinese prescription, has been widely used in the treatment of PCOS for decades. In previous studies, we found that SGD treatment could effectively reduce ovarian inflammation in PCOS rats. However, whether the anti-inflammation effect of SGD involves the regulation of the gut microbiota remains elusive. Methods: Letrozole-induced PCOS rat models were established, and the therapeutic effects of SGD were evaluated. Specifically, body weight, serum hormone concentrations, estrus phase and ovary histopathology were assessed. Then the structure of gut microbiota was determined by 16s rRNA sequencing. Additionally, the serum levels of pro-inflammatory cytokines and LPS were measured by ELISA kits. The key gene and protein expressions of TLR4/NF-κB signaling pathway were detected by quantitative real-time PCR and western blot. Results: SGD could effectively reduce body weight, regulate estrous cycles and ameliorate hyperandrogenism in PCOS rats. In addition, SGD treatment decreased releases of pro-inflammatory cytokines, enhanced the expressions of tight junction (occludin and claudin1), and then prevented a translocation of LPS into bloodstream. SGD could significantly reduce the ratio of Firmicutes to Bacteroidetes, decrease the abundance of LPS-producing pathogens Proteobateria and enrich the abundance of Butyricicoccus, Coprococcus, Akkermansia Blautia and Bacteroides in PCOS rats. Furthermore, SGD blunted the key gene and protein expressions of TLR4/NF-κB signaling pathway both in vivo and in LPS-induced RAW264.7 cells. Conclusion: SGD administration could ameliorate the inflammatory response in PCOS rats by remodeling gut microbiome structure, protecting gut barrier, and suppressing TLR4/NF-κB signaling pathway.

BACKGROUND: Patent foramen ovale (PFO) has been linked to the pathophysiology of cryptogenic stroke. Contrast transesophageal echocardiography (cTEE) is the current gold standard for PFO diagnosis, but it has the disadvantage of being semi-invasive and does not exempt from risks. As a diagnostic test, the efficacy of contrast transthoracic echocardiography (cTTE) and contrast transcranial Doppler (cTCD) is controversial. This study is aimed at investigating the efficacy of cTTE and cTCD versus cTEE in PFO detection, exploring a more cost-effective and reliable method for the diagnosis of PFO related to cryptogenic stroke. METHODS: From August 2019 to January 2020, a total of 213 patients with suspected PFO were included in our study. All patients underwent cTEE, cTCD, and cTTE examinations. cTTE3 was named for using a cutoff of 3 beats to detect PFO during cTTE, and cTTE5 represented a cutoff of 5 beats. A cutoff of cTCD grade III was named cTCD III. A cutoff of grade IV was named cTCD IV. cTTE3+cTCD IV was used for the combination of a cutoff of 3 beats during cTTE with grade IV of cTCD. cTTE5+cTCD III combined a cutoff of 5 beats during cTTE with cTCD grade III. Taking cTEE as the gold standard, we compared the sensitivity, specificity, negative likelihood ratio (-LR), and misdiagnosis rate for PFO detection among the above methods. RESULTS: A total of 161 of 213 (76%) patients had PFO confirmed by cTEE. With the spontaneous Valsalva maneuver, the sensitivity, specificity, negative likelihood ratio (-LR), and misdiagnosis rate of cTTE3 in PFO diagnosis were 60%, 90%, 44%, and 10%, respectively, and those for cTTE5 were 76%, 78%, 31% and 22%, respectively. The sensitivity, specificity, negative likelihood ratio (-LR), and misdiagnosis rate of cTCD III were 80%, 71%, 29%, and 29%, respectively, while those for cTCD IV were 55%, 90%, 49%, and 10%, respectively. When cTTE and cTCD were combined to diagnose PFO, the specificity and misdiagnosis rate were significantly improved, especially cTTE3+cTCD IV, with 100% specificity and a misdiagnosis rate of 0. CONCLUSION: cTTE or cTCD can be used for preliminary PFO related to cryptogenic stroke findings. The combination of the two methods can improve the specificity of PFO diagnosis, especially using the cutoff of cTTE3+cTCD IV.

The composite cathode material of the conductive polymer polyaniline (PANI)-coated spinel structural LiNi0.5Mn1.5O4 (LNMO) for high-voltage lithium-ion batteries has been successfully synthesized by an in situ chemical oxidation polymerization method. The electrode of the LNMO-PANI composite material shows superior rate capability and excellent cycling stability. A capacity of 123.4 mAh g-1 with the capacity retention of 99.7% can be maintained at 0.5C after 200 cycles in the voltage range of 3.0-4.95 V (vs Li/Li+) at room temperature. Even with cycling at 5C, a capacity of 65.5 mAh g-1 can still be achieved. The PANI coating layer can not only reduce the dissolution of Ni and Mn from the LNMO cubic framework into the electrolyte during cycling, but also significantly improve the undesirable interfacial reactions between the cathode and electrolyte, and markedly increase the electrical conductivity of the electrode. At 55 °C, the LNMO-PANI composite material exhibits more superior cyclic performance than pristine, that is, the capacity retention of 94.5% at 0.5C after 100 cycles vs that of 13.0%. This study offers an effective strategy for suppressing the decomposition of an electrolyte under the highly oxidizing (>4.5 V) and elevated temperature conditions.

BACKGROUND: Gorham-Stout disease, also known as vanishing bone disease, idiopathic massive osteolysis, is a rare entity of unknown etiopathology. This disease is characterized by destruction of osseous matrix and proliferation of lymphatic vascular structures and associated with massive regional osteolysis. It has a variable clinical presentation and is commonly considered as a benign disease with a progressive tendency and an unpredictable prognosis. The diagnosis is made by exclusion and based on combination with histological, radiological, and clinical features. Despite that several therapeutic options have shown certain efficacy, the effective treatment still remains controversial and there is no standard treatment to be recommended. CASE PRESENTATION: A previously healthy 40-year-old man presented with right lateral malleolus pain after an ankle sprain and was referred to our hospital. The radiographs indicated rapid massive bone destruction in the distal right lateral malleolus with an unclear margin. Based on the combination with histological, radiological, and clinical features, the diagnosis of Gorham-Stout disease was made. Considering that the residual function of malleolus had to be protected, prior bisphosphonate was used to control the progression of lesion, followed by surgical resection and biological reconstruction with autologous fibular bone grafting. The patient was followed up 8 years after surgery, he presented without progression and recurrence. CONCLUSIONS: We depict a case of Gorham-Stout disease at the right lateral malleolus and was successfully controlled by medication and surgical intervention. Based on the prior effective medical treatment, resection with biological reconstruction is a useful approach to treat Graham-Stout disease in bone.

PURPOSE: To investigate the survival of patients with primary osteosarcoma and lung metastases. METHODS: Clinical data of 128 patients with primary osteosarcoma and lung metastases who were admitted to Shenzhen Second People's Hospital from January 2013 to January 2015, were enrolled and retrospectively analyzed. Follow-up lasted for 3 years and after follow-up, patients were divided into survival group (n=54) and death group (n=74) according to their survival status. Univariate and logistic multivariate analyses were conducted on related factors contributing to death in patients with primary osteosarcoma and lung metastases. RESULTS: At the end of follow-up, there were 74 deaths (death group) and 54 survivors (survival group). Among all 128 patients, the 6-month, 1-year, 2-year and 3-year survival rates were 96.87, 92.96, 61.71 and 42.18%, respectively . Univariate analysis showed that tumor diameter, histological subtype, visceral pleural involvement, the number of lung metastatic lesions, the outcome after chemotherapy for the primary tumor, and therapy for lung metastasis were closely associated with death in patients with primary osteosarcoma and lung metastases (p<0.05). Multivariate logistic regression analysis showed that chondrocytes, multiple lung metastatic lesions, poor outcome after chemotherapy for primary tumor, and no surgery-combined treatment for lung metastasis were risk factors contributing to death in patients with primary osteosarcoma and lung metastases (p<0.05). CONCLUSION: Risk factors contributing to death in patients with primary osteosarcoma and lung metastases include chondrocytes, multiple lung metastatic lesions, poor outcome after chemotherapy for primary tumor, and no surgery-combined treatment for lung metastasis. Surgical resection and preoperative adjuvant chemotherapy can effectively improve the patient survival.

F-box proteins are essential components of the Skp-cullin-F-box complex (a type of E3 ubiquitin ligase), and participate in cell cycle and immune responses through the ubiquitin proteasome system. F-box protein 39 (FBXO39) belongs to the F-box family, which has been reported to be associated with cancer oncogenesis and progression. The present study aimed to investigate the role of FBXO39 in osteosarcoma (OS) cell proliferation and apoptosis in vitro. It was demonstrated that U-2OS cells exhibited high expression of FBXO39 compared with HOS and SaOS-2 osteosarcoma cells. Thus, knockdown of FBXO39 was performed using lentivirus-mediated short hairpin RNA (shRNA) transfection to validate the effect of FBXO39 in U-2OS cells. Western blotting and RT-qPCR analysis were used to confirm the efficiency of infection by analyzing the expression level of FBXO39. Using Celigo-based cell counting and MTT assays, it was demonstrated that FBXO39 knockdown significantly reduced the rate of cell proliferation compared with control. Caspase 3/7 activity assays and fluorescence-activated cell sorting confirmed the induction of apoptosis in U-2OS cells following FBXO39 knockdown. In conclusion, it was demonstrated that FBXO39 knockdown may significantly inhibit proliferation and promote apoptosis of U-2OS cells. Thus, FBXO39 may serve an important role in OS progression.

Programmed cell death-ligands 1 (PD-L1) is a key immune checkpoint protein and a promising therapeutic target for malignancy tumor immunotherapy. The prognostic value of PD-L1 in patients with bone and soft tissue sarcoma remains controversial. Therefore, this meta-analysis is conducted to evaluate the associations of PD-L1 expression with overall survival (OS), progression-free survival (PFS), and clinicopathological characteristics of sarcomaA comprehensive literature search of PubMed, Web of Science, Embase, and Cochrane Library was conducted for relevant studies. A total of 14 studies published from 2013 to 2017 were included. The pooled hazard ratios (HRs) with 95% confidence intervals (CIs) were extracted from included studies to assess the association between PD-L1 expression and OS, PFS of patients with sarcoma. Other relevant data were extracted to evaluate the correlations of PD-L1 expression with risk and clinicopathological characteristics of sarcoma. Stata 12.0 software was applied to calculate the strength of association between PD-L1 expression and sarcoma.In total, 14 articles containing 15 independent studies and 1,451 patients were included in this meta-analysis. We found that the high PD-L1 expression was associated with poorer overall survival (HR 1.27, 95% CI: 0.70-1.84 P = .000) and poorer events-free survival (HR 2.05, 95% CI: 1.55-2.70, P = .000) in bone and soft-tissue sarcoma patients. Additionally, we conducted subgroup analysis according to histology type, ethnicity, target of PD-L1 assessment, cutoff, the significant correlations with poor overall survival and events-free survival were also observed. In contrast none of the clinicopathological characteristics (gender, age, tumor site, tumor grade, tumor depth, tumor necrosis rate, metastasis, recurrence, chemotherapy, radiotherapy) was found to be associated with PD-L1 expression in our analysis.The findings from this meta-analysis indicate that PD-L1 expression might be a useful predicative factor of poor prognosis for patients with bone and soft tissue sarcoma.

A layered nanosphere structured NiO catalyst was successfully synthesized by a simple and efficient hydrothermal method as a cathode material for lithium-oxygen (Li-O2) batteries. Cyclic voltammetry (CV), dual electrode voltammetry (DECV) and chronoamperometry (CA) by rotating ring-disk electrode (RRDE) were carried out to investigate the catalytic activity of this catalyst for the oxygen evolution reaction (OER). The results revealed that the layered nanosphere NiO exhibited excellent electrochemical performance, stability and a typical four-electron reaction as a cathode electrocatalyst for rechargeable nonaqueous Li-O2 batteries. The overpotential of the NiO is only up to 0.61 V. X-ray photoelectron spectroscopy (XPS) characterization shows that the Li2O2 and Li2CO3 formed during the discharge process and decomposed after charging. Moreover, the cut-off voltage of discharging is about 2.0 V in the NiO-based Li-O2 batteries, while the specific capacity is up to 3040 mA h g-1. There is no obvious performance decline of the battery after 50 cycles at a current density of 0.1 mA cm-2 with a superior limited specific capacity of 800 mA h g-1. Herein, the layered nanosphere structured NiO catalyst is considered a promising cathode electrocatalyst for Li-O2 batteries.

Hereditary multiple exostosis (HME) is an autosomal inherited skeletal disease whose etiology is not fully understood. To further understand the genetic spectrum of the disease, we analyzed a five-generation Chinese family with HME that have observable inheritance. Exome sequencing was performed on three HME individuals and three unaffected individuals from the family. A downstream study confirmed a new C deletion at codon 442 on exon 5 of the exostosin-1 (EXT1) gene as the only pathogenic site which generated a stop codon and caused the truncation of the protein. We rediscovered the deletion in other affected individuals but not in the unaffected individuals from the family. Upon immunohistochemistry assay, we found that the EXT1 protein level of the patients with the novel mutation in our study was less than the level in the patients without the EXT1 mutation from another unrelated family. For a deeper understanding, we analyzed the mutation spectrum of the EXT1 gene. The present study should facilitate a further understanding of HME.
